prognosis of cancer. The aim of the present study is to investigate the possible influence of the
miR-499(rs3746444) polymorphism on the risk of gastric cancer in Chinese population. A total of
363 GC patients and 969 cancer-free controls were enrolled in this study. The genotypes were
obtained using MassARRAY method. The results showed that, compared with T allele, C allele was
associated with a significantly increased risk of GC (OR = 1.491, 95% CI = 1.155–1.923, P = 0.002).
Moreover, a significantly increased risk of GC in subjects with the TC genotype was observed
(adjusted OR of 1.559, 95% CI = 1.148–2.117, P = 0.004), compared with the wide type TT. We also
found that basically dominant model (TT vs. TC + CC) was suitable for the association between
rs6513497 and the risk of GC (OR = 1.568, 95% CI = 1.173–2.097, P = 0.002). However, the same
association was also shown in males and females. Meanwhile, rs3746444 was associated with the
tumor size of GC patients. The presentstudy indicated that miR-499 rs3746444 might contribute to

[Thésaurus Mesh]Tumeurs du rhinopharynxRésumé : Le cytochrome P450 2E1 (CYP2E1) est une enzyme de détoxication qui appartient à la phase I du métabolisme des xénobiotiques. Cette enzyme est codée par un gène très polymorphe dont le polymorphisme commun correspond à la substitution de la cytosine (C) par la thymine (T) en position –1019 (rs2031920). Ce polymorphisme a été défini dans plusieurs cancers notamment le cancer du nasopharynx (NPC). L’étude a porté sur 124 patients présentant un carcinome de nasopharynx, comparés à 166 témoins sains. La présence ou l’absence de ce polymorphisme est déterminée par la technique PCR-RFLP. La comparaison des fréquences entre les deux groupes est établie par le test χ2. L’analyse de nos résultats montre une différence significative entre les deux groupes concernant le génotype mutant (C2/C2) (5 % versus 0,5 % ; p = 0,04) et présente un facteur de risque pour le NPC en Tunisie (odds ratio = 8,39 ; IC 95 % [0,99–388,1]). Également, l’allèle C2 est associé significativement au groupe des patients plus que le groupe des témoins (6 % versus 2 %, p = 0,016) et augmente trois fois le risque de NPC en Tunisie (odds ratio = 2,99 ; IC 95 % [1,12–8,79]). Nos résultats confirment les résultats publiés dans les autres populations et mettent l’accent sur l’importance de l’implication de ce gène de détoxication dans la survenue du NPC, qui semble de plus en plus fortement associée à des facteurs environnementaux.

POLG is a gene that codes for the catalytic subunit of the mitochondrial DNA polymerase, which is involved in the replication of mitochondrial DNA. Genetic variants in mitochondrial DNA polymerase-γ (POLG) have been associated with several malignancies. However, as an important metabolic tissue, association between genetic polymorphisms of POLG and the prognosis and mitochondrial DNA (mtDNA) content in hepatocellular carcinoma (HCC) remains unknown. Here we investigated the association between in POLG with the prognosis and mitochondrial DNA (mtDNA) content in hepatocellular carcinoma (HCC).
Methods
Three nucleotide polymorphisms (SNPs) of rs1061316, rs2247233 and rs758130 in POLG were examined in 416 patients from two cohorts undergoing transcatheter arterial chemoembolization treatment. Leukocyte mtDNA content from 216 patients in cohort 2 was measured using a real-time PCR-based method. The association of SNPs with prognosis and of mtDNA content of patients was analyzed.
Results
The rs758130 in POLG gene was significantly associated with the prognosis of patients in a dose-dependent manner. Moreover, GG genotype in rs1061316 showed significantly high mtDNA content, an indicator of better prognosis.
Conclusions
Our study for the first time demonstrates that rs1061316 and rs758130 in POLG is associated with the prognosis and leukocyte mtDNA content in HCC patients.
